Cellomics HCS Reagent Kit for 3-parameter analysis of apoptosis.DescriptionKey criteria for determining whether a cell is undergoing apoptosis include distinct morphological changes in the appearance of the cell, as well as alterations in biochemical and molecular markers. The general patterns of apoptotic signals are similar, but the details of the pathways vary significantly depending on the cell type and apoptotic inducer. The Multiparameter Apoptosis 1 HCS Reagent Kit addresses the multiplicity of apoptotic pathways by providing High Content Screening (HCS)-qualified fluorescent reagents and a validated, optimized protocol for simultaneous measurement of three fundamental parameters associated with the process of apoptosis: (1) nuclear morphology, (2) mitochondrial mass and/or membrane potential, and (3) changes in f-actin content. Recognized as a hallmark of apoptosis, nuclear morphology (i.e. condensation or fragmentation) is measured using the Hoechst Dye. Loss of mitochondrial membrane potential and increases in mitochondrial mass have been recognized as early markers of apoptosis, and are quantifiable by MitoTracker® Red. Another indicator associated early in the apoptotic pathway is changes in the actin cytoskeleton, which are reflected by changes in Alexa Fluor® 488 Phalloidin staining f-actin. Prepared cells can be analyzed using standard fluorescence microscopy or using Cellomics’ fully automated HCS Readers, and the Cell Health Profiling Bioapplication affording automated plate handling, focusing, image acquisition, analysis, quantification, and data storage.
