Description
Cell cycle analysis is important for assessing potential cancer therapeutics. Current methods, including flow cytometry, require large numbers of non-adherent cells and present challenges for screening and automation. We have addressed these issues with the new Cell Cycle BioApplication. This flexible, multi-channel, High Content Screening (HCS) software module allows you to measure the cell cycle phase of individual cells and report population distributions, and to simultaneously correlate this with the presence or activation state of cell cycle-associated proteins in the same cell. Cell count and nuclear morphology are also reported.

A key benefit provided by the Cell Cycle BioApplication is the simultaneous analysis of three cell cycle-associated targets and the correlation of expression level or state of these targets with each other as well as with cell cycle phase. This is illustrated for DNA content and retinoblastoma protein in Figure 2.


The results obtained by this assay are very specific, and show excellent well-to-well reproducibility, as shown below in Figure 3. The application is further enhanced by including analysis of nuclear shape, which can be used to identify damaged, necrotic, or apoptotic cells from the normal cycling population. Cell Cycle also features reference well calculation, which provides the user with an automated approach for comparison of treated wells with a user-defined control population. The percentage of cells that are different compared to a control population are then reported as a well-level assay output feature. In summary, the Cell Cycle BioApplication delivers a flexible, multi-parameter tool for detailed analysis of cell cycle distribution for populations of adherent cells.
