Cellomics HCS Reagent Kit for measuring cell shape and size parameters.DescriptionCell attachment and spreading are multifactorial events mediated by interactions between adhesion molecules on the cell surface and underlying extra cellular matrices. Regulation of these events is central to a number of physiological processes including cancer cell metastasis, inflammation, and angiogenesis. Rather than measuring cell-ligand binding constants, this assay measures the ability of cells to respond by attaching and spreading on a specific substrate, providing a functional assay of the regulatory and structural molecules needed for morphological changes in the cell (Fig. 1). The efficacy of drugs that target the function or expression of these molecules can be measured by the rate of attachment and spreading on a tissue-specific substrate. The Cell Spreading HCS Reagent Kit provides high-content screening (HCS)-quality fluorescence reagents and a validated, optimized protocol necessary to quantify the spreading of cells by directly measuring their areas using a fluorescent cytoskeletal marker, rhodamine-conjugated phalloidin. The assay is performed on cells grown on standard high-density microplates. In addition to fluorescent phalloidin, the kit also includes the nuclear dye, Hoechst, enabling single cells to be distinguished from cell colonies or aggregates. A negative control compound, which prevents cells from spreading without affecting adherence to the underlying substrate, is also included in the kit. Prepared cells can be analyzed using standard fluorescence microscopes, or using a Cellomics fully automated HCS Reader with the Cell Spreading BioApplication, affording fully automated plate handling, focusing, image acquisition, analysis, quantification, and data storage. - Validated protocol for sample preparation/processing requires approximately 1.5 hours post-compound incubation
- Utilizes fluorescent reagents and requires no cell lysis, purification, or filtration steps; and no handling of radioactive isotopes
- Cells prepared with the HCS Reagent Kits may be analyzed directly via standard fluorescent microscopy; or using any HCS Imaging System
